In this study, ADC-derived interference was observed to substantially affect thelower limit of quantification (LLOQ) for the detection of free payloads, includingcommon commercial payloads such as monomethyl auristatin E (MMAE) andderuxtecan (DXD). To address this challenge, optimized LC-MS/MS methodswere developed, which effectively eliminated ADC-related interferences whileimproving LLOQ performance
