With the development of oligonucleotide drug, chemical modification and delivery system technology have made great progress, which increase the stability and targeting of oligonucleotide drugs, prolong the in vivo half-life, and improves drug efficacy. Liquid chromatography tandem mass spectrometry (LC-MS/MS) has high specificity and wide linear range, therefore can be used to for siRNA quantification in biological matrices for early discovery studies. In addition, liquid chromatography-high resolution mass spectrometry (LC-HRMS) can discover metabolites missed by other analytical methods, such as RT-qPCR and hybridization ELISA.
