Adenosine is an important signaling molecule and is composed of adenine and nucleoside. In hypoxic tumor microenvironment (TME), highly expressed CD73 catalyzes AMP dephosphorylation to form large amounts of adenosine. It can inhibit immune cell activity, enhance immune escape, and promote tumor growth. Traditional LC-MS method for adenosine is complicated, and sample derivatization, complex mobile phases (such as HILIC mode), or long analytical condition is required. This limits its application for research, particularly in studies with large number of samples.
The LeadSampler (LS-I) designed by Sound Analytics is built around a high speed sampling robot, with up to eight UHPLC capable valves that are seamlessly integrated to support multiple LC applications and workflows.
In this study, we developed a simple, direct, high throughput LS-I-LC-MS/MS method for the determination of adenosine. This will help scientists understand the role played by adenosine in TME and how it relates to cancer.
